Purpose:
: In the mature retina, Otx2 is expressed in three cell types: bipolar and photoreceptor cells that belong to the neural retina and retinal pigment epithelium (RPE), a neighbour structure that forms a tightly interdependent functional unit together with photoreceptor cells.
Methods:
we isolated the bone marrow stromal stem cells from pigmented rats and after reaching to passage 3-5, differentiated to neurosphere. The neurosphere medium changed to differentiated medium and in floating manner specific marker for progenitor cells (OTX2) and photoreceptor cell investigated.
Results:
our results showed that differentiated neurosphere 7-14 days after induction, express the retinal progenitor OTX2 marker and also Rhodopsin and recoverin, specific markers for photoreceptor cells.
Conclusion:
for the first time in vitro generation of OTX2 positive retinal progenitor cells create the hopeful for planning the cell replacement therapy for AMD and RP.
