Purpose:
|
Previous studies showed the anti-angiogenic effect of systemic propranolol. This study was designed to determine the safe dose of intravitreal propranolol.
|
Methods:
|
32 rabbits were randomized into 4 groups; 8 of which received intravitreal injection of 15µg (group B), 30µg (group C) and 60µg propranolol (group D) per 15µl and control group received 15µl normal saline (group A). Ophthalmic examinations and electroretinography (ERG) were performed at 1 and 4 weeks. After the last examination, animals were sacrificed and enucleated globes were processed for routine histopathology and glial fibrillary acidic protein (GFAP) immmunostaining.

Results:
|
ERG findings did not show any significant changes after intravitreal propranolol injection in groups A, B and C. However b wave amplitude significantly decreased in group D one week after intravitreal 60µg propranolol injection.(P=0.02) Histologic and GFAP tests confirmed ERG findings.

Conclusion:
|
This study demonstrated that intravitreal injection of 15µg and 30µg propranolol is safe in rabbit but intravitreal injection of 60µg propranolol is toxic.
